Title: IN VITRO PROPAGATION OF CHRYSANTHEMUM SPP USING SHOOT TIP AND SINGLE NODE EXPLANTS

Abstract: This study was carried out at Plant Tissue Culture of Kenena Center for Sugar Cane Research to evaluate the effect of different media formulation on morphogenesis of Chrysanthemum (pink and violet) Cultivars, shoot tip and nodal explants and growth regulator (auxins, cytokinins and their interaction). On MS medium morphogenesis of shoot tip and nodal explants of Chrysanthemum P was comparable, while on Chrysanthemum V. Shoot tip had significantly higher number of shoots and shoot growth on MS, but other parameters were similar on both explants. On B5 medium shoot tip was significantly higher on all morphogenetic parameters on both Chrysanthemum P and v except the number of shoots per explants. M S was better than B5 in number of shoots. BAP at 2.0mg/l was the best concentration for micro propagation of both nodal and shoot tip explants of Chrysanthemum P on MS medium, while BAP at 1.0 mg/l was the best concentration for propagation of Chrysanthemum v for shoot tip and nodal explants which were comparable in both genotypes. Kinetin at 0.5 mg/l induced significantly higher number of leaves, nodes and shoots from shoot tip and nodal explants of Chrysanthemum p and v the response of shoot tip was higher on shoot tip compared to nodal explants. Number of shoots induced by IBA was comparable on shoot tip and nodal explants of Chrysanthemum P except on MS medium which was significantly on Chrysanthemum v, the propagation of shoot tip was significantly better than nodal explants on all concentration. Number of shoots regenerated of Chrysanthemum p was significantly higher on MS compared to other NAA concentration on both nodal and shoot tip explants. However, plant height, numbers of leaves, number of Nodes were better on 0.25 mg/l on shoot tip of chrysanthemum p and comparable on all concentration when nodal explants were used. Number of shoot regenerated from shoot tip of Chrysanthemum v was significantly higher on MS. Number of leaves and plant height were better on 0.1 mg/l NAA. Rooting was induced on all concentration of NAA and IBA and the number of roots increase of auxins concentration
