Title: Isolation, Structure Elucidation, and Bioassy of Medicinal Plants from Alangasana

Abstract: This study describes the isolation, structural elucidation, chemical and biological screening of natural product from two species, Ozoroa insiginis (Anacardiaceae) and Ximenia americana (Olacaceae). The two species were selected for this study according to ethnobotanical literature and their significant traditional medicine in Alangasana area, in West of Sudan. The two species revealed high availability of tannins, saponins, steroids, flavonoids, and terpinoids. Alkaloids were present only in O. insiginis The antimicrobial activity of the ethanolic extracts from the barks of the two species was performed to determine quantitatively the presence or absence of inhibition zones and measurement of zone diameter. O.insiginis extract showed good activity against Pseudomonas aeruginosa, whereas X. americana extract exhibited significant activity against Staphylococcus aeruginosa and low activity against Pseudomonas aeruginosa. Two novel compounds were isolated from column fractions of O. insiginis extract, SB1: (6-tetradecyl-1,6-diazabicyclo[3.2.0]hepta-2,4dien-7-one) was obtained from the 2nd column, FII,. It showed a characteristic colour of alkaloids on TLC plate when it was sprayed with Dragindorff,s reagent. The second compound SB8 was an alkaloid, and was identified as: 3-hydroxy-1-tridecyl-1H-pyrrolo [1, 2-α] imidazole-2-carboxylic acid.It was obtained from the 1st column, F1. SB8 have shown high cytotoxicity against brine shrimps. Four compounds were isolated from the ethanolic extract of the bark of X. americana species, two of them were new and the other two were popular. One of the two new compounds SC2 is: 17-( 1- (3-isopropyle-3- metyloxiran-2-yl)propan-2-yl)-10,13-dimethyl-6,7,8,9,10,11,12,13,14,15,16,17-dodecahydro-cyclopenta[α]phenanthren-3(2H)-one. It exhibited a characteristic colour of steroids. The other new compound was SC9: 3- Methyl-1-oxoiso chroman-8-Carboxylic acid. Those two compounds SC2 and SC9 were characterized, and by the application of Science Finder unit of Hussein Ebrahim Jamal institute (HEJ), Karschi University, it has been shown that they were the first reported compounds isolated fromX.americana species. The two popular steroidal compounds that were isolated from this species, one was SC5: Stigmast-5-en-3-ol. It showed high cytotoxicity towards the brine - shrimps. Compound SC11: Ergosta- 4, 6, 8, 22-tetraen-3-one, was fluorescent.
