Title: Laboratory Evaluation of Some Disinfectants Used in Poultry Farms Against Some Bacterial Isolates

Abstract: This study was designed to evaluate the effect of the four disinfectants: Quaternary Ammonium Compound QAC (Vrocid), Iodine compound (Iocid 30), Formaline (Aldekol des o3) and H2O2 (Aquaclean) used by commercial poultry integrator against four bacterial isolates; Escherichia coli(E.coli), Staphylococcus aeureus, Proteus spp. and Pseudomonas spp.and the effect of the heat in relation to time on recommended concentration. This experiment consist of two-phases trials; in the first part these disinfectants were made in five concentrations (QAC: 0.15%, 0.25 %, 0.35%, 0.45%, 0.55%), (Iodine: 0.15%, 0.25%, 0.35%, 0.45%, 0.55%), (Formalin: 0.4%, 0.5%, 0.6%, 0.7%, 0.8%) and (H2O2:9%, 10%, 11%, 12%, 13%), one according to the manufacturer’s recommendation (QAC:0.25 %), (Iodine: 0.25%), (Formalin: 0.5%), (H2O2:10%) three were higher (QAC:0.35%, 0.45%, 0.55%), (Iodine: 0.35%, 0.45%, 0.55%), (Formalin: 0.6%, 0.7%, 0.8%), (H2O2:11%, 12%, 13%) and one was lower (QAC: 0.15%), (Iodine: 0.15%), (Formalin: 0.4%), (H2O2:9%) than the manufacturer recommendation, these concentrations were made in disk from filter paper. Five concentrations of each disinfectant were kept in a plate that contained the bacterial culture. The disc which was saturated by the recommended concentration of each disinfectant was put in the middle of the plate containing bacterial isolates and the lower and higher concentration was rounded. The plates were incubated at 37oC overnight and then observed, the diameter of clear inhibition zone surrounding each disc was measure using a ruler.In the second part,only the recommended concentration of each disinfectant (QAC:0.25 %), (Iodine: 0.25%), (Formalin: 0.5%), (H2O2:10%) was used, each disinfectant was divided in four groups and submerged in a water bath at 37, 45, 50, 60oC, each group contained 4 tubes. The tubes were collected at 5, 10, 15, 30 minutes after the start of the experiment, the disk was put inside the tube of each disinfectants then taken immediately and was put in the plate containing bacterial isolates which were divided in 4 departments. Incubated overnight at 37oC, then the plates were observed, the diameter of clear inhibition zone surrounding each disk was measured using a ruler. The laboratory evaluation indicated that H2O2(E.coli:13%, staphylococcus aureus: 11%, Proteusspp:10% and Pseudomonasspp 10%)QAC (E.coli:0.35%, 0.45% and 0.55%, staphylococcus aureus: 0.55%, Proteusspp:0.15%,0.25% and 0.45% and Pseudomonasspp 0.55%) and Formaldehyde (E.coli:0.8%, staphylococcus aureus: 0.5% Proteusspp:0.4% and Pseudomonasspp 0.6%) respectively were effective against E.coli, Staphylococcusaureus and Proteus spp. and pseudomonasspp.while the iodine did not show any effect. It was concluded that the disinfectants used in this study were very effective and recommended to be used in poultry houses
