Title: Validity of Eijkhman's Test for Species of the Genus Escherichia

Abstract: Eijkhman's test it is a mean of separating E . coli of fecal origin that ferment lactose at 44ºC from other E . coli strains as stated by the American Puplic Health Association (1925) . The objective of this study was to isolate different species of the genus Escherichia from various sources and study their behavior towards Eijkhman's test. A total of (120) samples were collected. Fifty samples were collected from the contents of the small intestines of calves from various slaughterhouses. Thirty samples were collected from the small intestines of sheep from the slaughtered animals in various abattoirs. Twenty five samples were collected from beef and lamb vended meat from Omdurman market and fifteen samples were collected from the gut of the cockroach (Periplaneta americana). Sixty four (64) isolates from the total one hundred and twenty samples were identified as Escherichia spp . based on their morphology and biochemical characters the species were identified as: E. coli, E. fergusonii, E. vulneris, E. albertii, E. blattae and E. adecarboxylate. Eijkhman's test was applied for all the identified species. They were incubated at 44ºC in a water bath and read after 48 hours . It was found that E. coli (80%), E. fergusonii (50%) E. vulneris (20%) and E. adecarboxylate (70%) were positive for the test due to fermentation of lactose at 44ºC and production of gas that filled Durham’s tube. While this is so, E.albertii and E. blattae were lactose negative that is why they were eleminated from Eijkhman’s test. Further studies are needed to test more strains of the species of the genus Escherichia for Eijkhman’s test as well as the new emerging species.
