Title: TOXICOPATHOLOGICAL EFFECTS OF THE EXTRACTS OFAMBROSIA MARITIMA AND GUIERA SENEGALENSIS IN RATS

Abstract: The toxicity of the methanolic and water extracts of Ambrosia maritima and Guiera senegalensis for rats treated for three weeks at different doses was determined. The methanolic extract was injected intramuscularly and the water extract was given orally. A single intramuscular dose of ٢٠٠٠mg/kg of Ambrosia maritima methanolic extract was fatal but a daily dose of ٥٠٠and ٢٥٠mg/kg caused inappetence, decreased activity, lameness and even paralysis although body weights were not affected. The main lesions of the toxicity were pale enlarged livers, enlarged spleens and dry gangrenous muscles. Diarrhoea was the most prominent sign in rats receiveing Ambrosia maritima water extract at the doses ١٠٠٠ and ٥٠٠mg/kg while at the dose ٢٥٠mg/kg no clinical signs were observed. Lesions in both extracts consisted of generalized congestion, enterohepatonephropathy and pancreatic hyperplasia. Organ lesions were accompanied by leucocytosis and alteration of serum alkaline phosphatase activity, cholesterol level and concentration of total protein, calcium and inorganic phosphorus. Clinical signs of Guiera senegalensis methanolic extract were not clear. Rats receiving ١٠٠٠ and ٥٠٠mg/kg died within one week. The dose ٢٥٠mg/kg resulted in enlarged livers and kidneys. Alteration of haematological and serological parameters were evident. The water extract of Guiera senegalensis caused less marked effects. Endotheliotoxicity, hepatonephropathy and pancreatic hyperplasia characterized the treatment with Guiera senegalensis.
