Isolation, Characterization and Nematocidal Activity of Bacillus Thuringiensis Isolated From Soil and Water

Abstract
In this study 80 soil samples and 20 water samples were collected from eight different localities in Khartoum state with the objective of isolation and characterization of Bacillus thuringiensis strains having nematocidal activities. The samples were subjected to selective heating at 80=616;C for ten minutes to kill all vegetative cells .Using different morphological and biochemical tests seven strains out of 106 Bacillus isolates were identified as Bacillus thuringiensis belonging to five various subspecies . Two isolates belonged to subspecies sotto serotype H-4, two belonged to subsp israelensis serotype H-14 and the three remaining isolates belonged to subsp thuringiensis serotype H-1, subsp aizawai serotype H-7 and subsp kurstaki serotype H-3. The seven strains of B.thuringiensis were tested for their pathogenic effect on three plant parasitic nematodes namely Radopholus similis, Meloidogyne sp and Ditylenchus dipsaci. Some strains such as Sh24,K94 and H102 resulted in the death of all of the nematodes (i.e.100% mortality) whereas D35,S50,U68 and T77 caused mortality percentages that ranged from 30 – 95.This study clearly demonstrated the nematocidal activities of the indigenous B.thuringiensis strains.
