Interleukin-10 (IL-10) gene polymorphisms in development of Post Kala-azar dermal leishmaniasis (PKDL)

goal of this study to determine if there is an association of IL-10 promoter gene polymorphisms with Post Kala-azar dermal leishmaniasis (PKDL) in the highly susceptible Masalit tribe from three neighboring villages Um-Salala, EL-Rugab and Ali babkir. DNA was collected from 85 families consist of 51 trios and 34 pairs. - 592C/A, - 819C/A and -1082A/G of IL-10 promoter gene polymorphisms were genotyped using PCR-RFLP and ARMS-PCR techniques. The data were tested to their fit to Hardy- Weinberg Equilibrium and the polymorphisms were analyzed using Transmission Disequilibrium Test (TDT) and Haplotype Based Score Test to test the association to the disease. All polymorphisms were in Hardy-Weinberg Equilibrium. TDT analysis showed No evidence of association between IL-10 -592C/A, -819C/T and -1082A/G and susceptibility to PKDL (p>0.05, 0.7855, 0.7815, and 0.5862, respectively). Haplotype Based Score Test showed border significant between -592C/T and -1082A/G (p=0.053). All eight haplotype probabilities of three target loci were observed in Masalit tribe with higher frequency of GCC and ATA haplotype 32% and 30% respectively
